The aquaporin (AQP) family comprises orthodox AQPs that transport water, glycerol, and H2O2, as well as unorthodox AQPs displaying the Asn-Pro-Cys (NPC) motif, which confers a unique capacity to transport hydrogen peroxide. In the brain, AQP11 is an archetypical member of this family and can be detected in the choroid plexus, blood-brain barrier (BBB), and cerebral white matter. However, its role in the brain remains to be elucidated.

Immunocytochemistry (ICC) showed that AQP11 was primarily localized at the epithelium of the choroid plexus and at the endothelium of the brain capillary. Its staining partially overlapped with that of glucose transporter 1 (GLUT1), a marker for the capillary endothelium. In contrast, AQP11 expression was not observed at astrocyte endfeet or glomerules of the kidney and liver, suggesting that it may act in parallel with AQP4 to transport water across the BBB.

In astroglial and neuronal cell lines, LPS stimulation significantly increased AQP0 and AQP11 protein levels, but not transcripts (Figure 4E-H). The expression of AQP0 and AQP11 in cells exposed to basal and LPS-stressed conditions was further evaluated by indirect immunoperoxidase assay.
